Description

GlykoPrepHCcartridgesaresolid-phaseextractionspincartridges,packedwith~5mgofHypercarb™porousgraphiticcarbon(PGC),designedforpurificationofglycansfromnon-carbohydratematerial,includingsaltsandproteins.Whensamplesareloadedonthecolumn,saltspassthrough,whileglycansandproteinadsorbontothematrix.Theglycansmaythenbeselectivelyelutedfromthecolumn.

Applications:

• DesaltingofN-glycansreleasedbynativedigestionwithe.g.PNGaseF
• Proteinremovalanddesaltingfollowingexoglycosidasedigestionofreleasedglycans(e.g.sialidasedigestion,fucosidasedigestionpriorto2-ABlabeling)
• Cleanupofglycansreleasedbyhydrazinolysis
• Desaltingglycansamplesincludinganionicspeciespriortomassspectrometry
• Cleanupoffluorescently-labeledglycans(e.g.glycanslabeledwith2-AB,InstantAB™)fromlabelingreactions*

Note:thesecartridgesarenotsuitableforremovingdetergent.

*NotrecommendedforcleanupofInstantPC-labeledN-glycans,asthesemustbestoredinGly-X/InstantPCEluentwith10%ACN.SamplesappliedtoHCcartridgesmustbeaqueousorloworganic(<5%).

Sampleprepconsiderations:

•Glycansamplestobecleanedmustbeinanaqueousbufferorabuffercontainingalowpercentage(<5%)oforganicsolvent.
•Ifthesamplecontainsorganicsolvent,dilutewithwateruntiltheorganicsolventcontentislessthan5%byvolume.Forexample,dilutea5μl2-ABlabelingreactionwith95μlwaterpriortoloADIng.
•SamplesshouldnotcontaindetergentssuchasSDS.
•SuggestedstartingupperlimitfortheHCcartridgesis~50μgglycans.Thebindingcapacityforspecificglycoproteins&glycansmayneedtobedetermined.Minimumglycanloadwilldependonthesensitivityoftheanalyticalmethodsused.

TheHCleanupModulerequiresminimalequipment.Allyouneedisacentrifugewithadeep-wellplaterotor(minimum44mmheight)andaStarterLabwareKit(AM200).

Alternatively,thecartridgesmayberuninamicrocentrifugeusingEppendorftubesandAM400adapters-pleaseseetheUserManualfordetails.
